چکیده

The present study was concerned with the selection of suitable surfactant for the production of alpha amylase by Bacillus subtilis GCBM-25 in 250 ml shake flask containing 50 ml of fermentation medium. Different surfactants (laundry soap, detergent powder, sulphonic acid, acyle benzene sulphonic acid, liquid soap, Tween 80, sodium silicate, bath soap, sodium tripolyphosphate, sodium lauryl ether sulphate or sodium lauryl sulphate) @ 2.0 % (w/v) were tested for enzyme production. Of all the surfactants, laundry soap gave better production of alpha amylase (605 U/ml/min) 44 h after inoculation (4.0 % inoculum size). The production of enzyme was found to be optimum (857 U/ml/min) when Millon soap @ 3.2 % (w/v) was added to the medium. The thermostability of the enzyme was decreased from 70 to 50C as the surfactant was added to the fermentation medium.

Bacillus Amyloliquefaciens Alpha-amylase Inhibition by Organic Solvents: A Study on Methanol, Ethanol and Propanol

Alpha-amylase is widely used as an industrial enzyme, and a therapeutic target for which inhibitors are designed. Organic solvents are used to dissolve various compounds that would be studied as moderators of alpha-amylases, but they could themselves affect enzyme activity and stability. Methanol, ethanol and propanol are simple alcohols that may be commonly used to this end, and their effect h...

Comparison of the alpha-amylase of Bacillus subtilis and Bacillus amyloliquefaciens.

The alpha-amylase (alpha-1,4-glucan 4-glucanohydrolase, EC 3.2.1.1) of Bacillus subtilis strain W23 is less negatively-charged than the alpha-amylase of B. amyloliquefaciens strain F, as determined by electrophoretic mobility in polyacrylamide gel at pH 8.6. The alpha-amylase of strain W23 is immunologically unrelated to the alpha-amylase of strain F, as judged by lack of cross-reaction in Ouch...

NOTES Partial purification and some properties of α-amylase from Bacillus subtilis KIBGE-HAS

An extracellular α-amylase from Bacillus subtilis KIBGEHAS was partially purified by ultrafiltration and ammonium sulphate precipitation with 19.2-fold purification and specific activity of 4195 U/mg. The enzyme showed relatively high thermostability and retained 62% of its activity when kept at 70°C for 15 min. α -Amylase was highly stable at −18°C and loss of activity was very low during stab...
